DLL3 CAR-T is a chimeric antigen receptor (CAR)-modified T cell therapy designed to target Delta-like 3 (DLL3), a protein highly expressed on small cell lung cancer (SCLC) cells with minimal expression on normal adult tissues. The CAR construct utilizes the SC16.15 single-chain variable fragment (scFv) for DLL3 targeting, combined with CD8α hinge and transmembrane regions, and intracellular signaling domains from CD28, 4-1BB, and CD3ζ, representing a third-generation CAR format. This cell-based immunotherapy aims to engage and activate T cells to efficiently kill DLL3-positive cancer cells, offering a potential therapeutic approach for SCLC.
