DLL3 x B7H3 bispecific ADC is a novel bispecific antibody-drug conjugate (ADC) developed by GeneQuantum Healthcare, designed to target both Delta-like ligand 3 (DLL3) and B7-H3 (CD276). This therapeutic candidate is engineered to address the significant molecular heterogeneity and treatment resistance observed in small cell lung cancer (SCLC). DLL3 is an inhibitory ligand of the Notch pathway that is highly expressed in neuroendocrine SCLC subtypes (SCLC-A and SCLC-N), while B7-H3 is homogeneously overexpressed across all SCLC subtypes, including the chemo-immunotherapy resistant SCLC-P variant. The ADC utilizes a topoisomerase 1 inhibitor (Top1i) payload conjugated to a bispecific antibody format. Preclinical data presented at AACR 2026 demonstrated that the lead candidate exhibits potent cellular internalization and target-specific cytotoxicity, with superior antitumor activity in patient-derived xenograft (PDX) models compared to monospecific ADC benchmarks, particularly in refractory SCLC-P models.
