DLL3xB7H3 bsADC is a preclinical-stage bispecific antibody-drug conjugate (bsADC) developed by Biocytogen. It is designed to simultaneously target two tumor-associated antigens, Delta-like ligand 3 (DLL3) and B7-H3 (CD276), which are frequently co-expressed in small cell lung cancer (SCLC) and other neuroendocrine tumors. The therapeutic consists of a bispecific antibody in a 1+1 format conjugated to a novel topoisomerase I (TOPO1) inhibitor payload with a drug-to-antibody ratio (DAR) of 8. The conjugation utilizes a highly hydrophilic, enzyme-cleavable linker intended to provide a potent bystander effect while maintaining stability. This dual-targeting strategy aims to enhance tumor selectivity, address antigen heterogeneity, and improve efficacy over monospecific ADCs. Preclinical studies have demonstrated superior anti-tumor activity in various models compared to existing benchmarks, suggesting potential as a first-in-class treatment for SCLC.
