The DLL3xCD3xCD2 trispecific antibody is a novel T-cell engager (TCE) designed to target Delta-like ligand 3 (DLL3) on tumor cells while simultaneously engaging CD3 and the CD2/CD58 co-stimulatory pathway on T cells. Developed by Probio, Inc., this therapeutic candidate utilizes an internally developed CD3 VHH antibody (nanobody) with cynomolgus cross-reactivity. By integrating a CD2 co-stimulatory moiety, the antibody aims to enhance T-cell activation, proliferation, and persistence within the immunosuppressive tumor microenvironment (TME), addressing common limitations of conventional bispecific TCEs such as T-cell exhaustion and insufficient infiltration. Preclinical studies in human PBMC-reconstituted CDX models of small cell lung cancer (SCLC), including SHP77 and NCI-H82, have demonstrated superior antitumor efficacy and a balanced cytokine release profile compared to clinical benchmarks like tarlatamab. The molecule is currently undergoing IND-enabling studies for the treatment of solid tumors.
