DLST RNAi refers to the use of RNA interference (RNAi) to suppress the expression of dihydrolipoamide S-succinyltransferase (DLST), the E2 component of the alpha-ketoglutarate dehydrogenase complex (KGDHC). DLST plays a vital role in the tricarboxylic acid (TCA) cycle by converting alpha-ketoglutarate to succinyl-CoA. Research presented at AACR 2016 demonstrated that DLST is essential for the survival and pathogenesis of MYC-driven T-cell acute lymphoblastic leukemia (T-ALL). Knockdown of DLST in human T-ALL cell lines leads to metabolic disruption, including the accumulation of alpha-ketoglutarate and glutamine, resulting in decreased cell viability and the induction of apoptosis. This metabolic dependency suggests that DLST is a potential therapeutic target for MYC-mediated hematological malignancies.
