DLYE5953A is an antibody-drug conjugate (ADC) targeting the tumor-associated antigen lymphocyte antigen 6 complex locus E (Ly6E). It consists of a monoclonal antibody directed against Ly6E conjugated to a cytotoxic payload. Ly6E is overexpressed in various solid tumors including breast, lung, gastric, ovarian, pancreatic, kidney and head/neck carcinomas. Upon intravenous administration, the ADC binds to Ly6E on tumor cells and is internalized via endocytosis; this leads to lysosomal accumulation and subsequent release of the cytotoxic agent inside cancer cells. Preclinical studies demonstrated inhibition of cell proliferation and durable tumor regression in LY6E-expressing xenograft models. In clinical trials (Phase I), DLYE5953A showed acceptable safety and preliminary antitumor activity in patients with advanced or metastatic solid malignancies that had progressed on standard therapy[1][2][4][7].
