DM-5689 (also known as DS-5689) is a small molecule agonist of the G protein-coupled receptor 119 (GPR119), which was developed by Daiichi Sankyo for the treatment of type 2 diabetes mellitus and obesity. GPR119 is a G protein-coupled receptor primarily expressed in the pancreatic beta cells and the enteroendocrine cells of the gastrointestinal tract. By activating GPR119, DM-5689 stimulates the secretion of insulin from the pancreas and the release of incretin hormones, such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP), from the gut in a glucose-dependent manner. This dual mechanism of action aims to improve glycemic control and promote weight loss. DM-5689 reached Phase 1 clinical trials, where its safety, tolerability, and pharmacokinetics were evaluated in healthy volunteers and patients with type 2 diabetes. However, development of the compound was subsequently discontinued, and it is no longer listed in the developer's active pipeline.
