DM1157 is a novel antimalarial drug candidate derived from chloroquine. It is designed to overcome drug resistance in malaria parasites by inhibiting the formation of hemozoin, which protects Plasmodium falciparum from heme toxicity. The drug also binds to the Plasmodium falciparum chloroquine resistance transporter (PfCRT), potentially impairing drug efflux and overcoming common resistance mechanisms. However, a Phase 1 clinical trial was halted early due to significant toxicity observed in healthy adult volunteers.
