DM919 is a humanized IgG1 monoclonal antibody developed by D2M Biotherapeutics for the treatment of advanced solid tumors. It specifically targets MHC class I polypeptide-related sequence A and B (MICA/B), which are stress-induced ligands highly expressed on cancer cells. These ligands are recognized by both innate and adaptive immune cells via NKG2D receptors. Tumor cells evade immune detection by shedding MICA/B from their surface, thereby escaping NKG2D-mediated immune surveillance. DM919 blocks the shedding of MICA and MICB proteins from cancer cells, stabilizes their expression on the cell surface, and presents shed MICA/B to activate NKG2D on NK or T cells, restoring and enhancing anti-tumor responses mediated by T-cells and natural killer (NK) cells. The drug also mediates antibody-dependent cellular cytotoxicity (ADCC) against tumor cells. Preclinical studies have shown significant antitumor activity as a single agent across various tumor models, with additional synergistic effects when combined with anti-PD1 therapy such as pembrolizumab[1][2][4][5][6].
