**DM919 + pembrolizumab** is an investigational combination therapy for advanced solid tumors, currently in Phase 1 clinical trials. DM919 is a humanized monoclonal antibody targeting MICA/B proteins, which are stress-induced ligands highly expressed by cancer cells. DM919 inhibits the shedding of MICA/B from tumor cells, restoring and enhancing NKG2D-dependent activation of T and NK cells to promote anti-tumor immune response. Pembrolizumab is a monoclonal antibody that blocks the PD-1 receptor on T cells, preventing tumor-mediated immune evasion and further amplifying the immune response. The combination aims to synergize both innate (via DM919) and adaptive (via pembrolizumab) immune responses, potentially overcoming resistance in tumors with low MHC-I expression. The combination is being investigated to establish safety, tolerability, pharmacokinetic/pharmacodynamic profiles, and preliminary efficacy in patients with advanced or metastatic solid tumors[1][2][3][4][6].
