DMD06-MAB is a monoclonal antibody currently in development for the treatment of Duchenne muscular dystrophy (DMD). Developed through a collaboration between Ospedale San Raffaele and the University of Manchester, this therapeutic candidate targets the P2X purinoceptor 7 (P2RX7), a purinergic receptor that acts as an ATP-gated ion channel. In the context of DMD, the muscle microenvironment contains abnormally high levels of extracellular ATP due to chronic muscle fiber damage. This leads to the overactivation of P2RX7 on muscle cells and infiltrating immune cells, triggering pro-inflammatory cytokine release, oxidative stress, and further muscle cell death. By antagonizing the P2RX7 receptor, DMD06-MAB aims to interrupt this pathological cycle, thereby reducing chronic inflammation, limiting muscle fiber necrosis, and potentially preserving muscle function in patients with DMD.
