Overview

The DMF5 TCR retroviral vector (also designated MSGV1-F5AfT2AB) is a gamma-retroviral gene therapy vector developed at the Surgery Branch of the National Cancer Institute (NCI). It is designed to genetically engineer autologous peripheral blood lymphocytes (PBLs) to express a high-affinity T-cell receptor (TCR) that recognizes the MART-1(27-35) epitope (AAGIGILTV) in the context of HLA-A*0201. The TCR was originally cloned from a tumor-infiltrating lymphocyte (TIL) clone, DMF5, isolated from a patient with metastatic melanoma. This adoptive cell therapy approach redirects the patient's T cells to specifically target and lyse melanoma cells expressing the MART-1 antigen (Melan-A). The vector has been utilized in clinical trials to treat patients with advanced metastatic melanoma, demonstrating the ability to generate polyfunctional CD4+ and CD8+ T cells capable of cytokine secretion and direct tumor cell killing.
