dMIP3α-gp100 is an experimental DNA vaccine composed of a fusion between the chemokine MIP3α (also known as CCL20) and the melanoma-associated antigen gp100. This construct is designed to target and deliver the gp100 tumor antigen directly to immature dendritic cells via the MIP3α component, which binds to CCR6 on dendritic cells, thereby enhancing antigen presentation and the subsequent T-cell immune response. Preclinical studies in the B16F10 mouse melanoma model show that this chemokine-antigen fusion vaccine enhances immunogenicity, increases vaccine-specific CD8+ tumor-infiltrating lymphocytes, and improves survival compared to antigen-only approaches. The vaccine platform is modular and potentially adaptable for various antigens and indications, notably for therapeutic cancer vaccines and neoantigen personalized therapies[1][7].
