Overview

DMPA-SC (subcutaneous depot medroxyprogesterone acetate) is a long-acting, progestin-only injectable contraceptive administered subcutaneously. It contains 104 mg of medroxyprogesterone acetate in a depot formulation and provides contraception for three months per injection. The mechanism of action involves suppression of ovulation by inhibiting the secretion of gonadotropins (luteinizing hormone and follicle-stimulating hormone), thickening cervical mucus to prevent sperm penetration, and thinning the endometrial lining to inhibit implantation. DMPA-SC is available as an all-in-one prefilled injection system (notably Sayana Press) that can be self-administered or given by healthcare providers. It is indicated for prevention of pregnancy and management of endometriosis-associated pain[2][4][5][8][9].
