DMR-001 is a preclinical, Fc-null monoclonal antibody that selectively targets mutant calreticulin (mutCALR), a pathogenic driver mutation in myeloproliferative neoplasms (MPNs) such as essential thrombocythemia and myelofibrosis. Engineered by Damora Therapeutics and now being advanced by Galecto following its acquisition of Damora, DMR-001 has demonstrated enhanced binding affinity and approximately tenfold higher potency against type 2 mutCALR compared with a reference antibody in preclinical studies, and incorporates half-life extension technology to enable low-volume, infrequent subcutaneous dosing.[5][7][9][11][13] By selectively neutralizing mutCALR-driven signaling on hematopoietic cells, DMR-001 is positioned as a potentially best-in-class, disease-modifying targeted therapy within an emerging class of mutCALR-directed biologics for MPNs, with an IND submission and first-in-human subcutaneous administration planned in the mid‑2020s.[7][11][14][15]
