Overview

DMRA-101 is a preclinical biologic candidate, specifically a monoclonal antibody, being developed by Damora Therapeutics for the treatment of myeloproliferative neoplasms (MPNs), including essential thrombocythemia and myelofibrosis. The drug is designed to selectively target mutant calreticulin (CALR), a protein that, when mutated, acquires a unique C-terminal tail. This mutant tail constitutively binds to and activates the thrombopoietin receptor (MPL), driving the overproduction of blood cells characteristic of MPNs. By binding to the mutant CALR protein, DMRA-101 aims to block this pathogenic interaction and normalize MPL signaling. Damora Therapeutics is currently advancing DMRA-101 through IND-enabling studies, with an Investigational New Drug (IND) filing anticipated in 2026.
