Overview

DMRA-201 is a preclinical biologic candidate, specifically a monoclonal antibody, being developed by Damora Therapeutics for the treatment of myeloproliferative neoplasms (MPNs). It is designed to target mutant calreticulin (CALR), a driver mutation found in approximately 30% of patients with essential thrombocythemia and primary myelofibrosis. Mutant CALR proteins possess a unique, positively charged C-terminal neoepitope that constitutively activates the thrombopoietin receptor (MPL), leading to oncogenic signaling. DMRA-201 aims to selectively bind this mutant protein to inhibit the pathogenic MPL activation and potentially induce the clearance of malignant hematopoietic stem cells. The developer expects to file an Investigational New Drug (IND) application for its CALR-targeting program in 2026.
