Overview

DMXB-A (also known as GTS-21 or DMBX-anabaseine) is a synthetic small molecule and derivative of anabaseine. It acts as a selective partial agonist at the alpha7 subtype of nicotinic acetylcholine receptors (nAChRs), with much weaker activity at the alpha4beta2 subtype. By activating alpha7 nAChRs in the brain, it has demonstrated neuroprotective and cognitive-enhancing effects in preclinical models and early clinical trials. Investigational indications have included Alzheimer's disease, schizophrenia, Parkinson's disease, attention-deficit hyperactivity disorder (ADHD), autism spectrum disorder, nicotine dependence/smoking cessation support, and rheumatoid arthritis. Clinical development reached Phase II for several indications but was discontinued or withdrawn before approval[1][2][5][6].
