DN-104 is a next-generation therapeutic program targeting Carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5), developed by DAAN Bio Therapeutics. The program utilizes a monoparatopic antibody designed with low affinity for soluble CEACAM5, a strategy intended to minimize off-target sequestration by circulating antigens and enhance tumor-specific localization. DN-104 is being advanced through two distinct therapeutic modalities: an antibody-drug conjugate (ADC) in partnership with LigachemBio (where it is designated as LCB58A) and a CAR-T cell therapy in partnership with GC Cell. Both programs are aimed at treating CEACAM5-positive solid tumors. As of early 2025, both programs have been licensed out, with the ADC program anticipated to reach significant development milestones by approximately 2027.
