Overview

DN1406131 is a selective, orally administered small molecule dual inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase 2 (TDO2). Both IDO1 and TDO2 catalyze the first and rate-limiting step in the conversion of tryptophan to kynurenine. By inhibiting these enzymes, DN1406131 blocks production of kynurenine—a ligand for the aryl hydrocarbon receptor (AhR)—thereby disrupting an immunosuppressive pathway that tumors exploit to evade immune surveillance. The drug is being developed primarily as an antineoplastic agent for solid tumors[1][2][3].
