Overview

DN604 is a **platinum(II) carboplatin analogue** designed with a functional dicarboxylato ligand, investigated as a novel antitumor agent. It demonstrates potent in vitro and in vivo efficacy, particularly in **gastric cancer cells (SGC-7901)** and shows activity comparable to cisplatin but with reduced toxicity. The mechanism involves inhibition of **CK2-phosphorylated cdc25C**, leading to decreased activation and altered subcellular localization of p-cdc25C, inactivation of cdc2/Cyclin B, and subsequent G2/M cell cycle arrest and apoptosis in cancer cells. DN604 thus exploits disruption of checkpoint proteins to inhibit tumor growth. Additionally, it has shown effectiveness in preclinical models against cisplatin-resistant tumor cells with less toxicity compared to standard platinum drugs[1][3].
