Overview

The DNA + HIV-rAd5 vaccine is an investigational preventive HIV-1 vaccine regimen that uses a "prime-boost" strategy. The priming component consists of a multiclade plasmid DNA vaccine encoding several HIV-1 proteins (Gag, Pol, Nef from clade B and Env from clades A, B, and C), followed by a boosting component using recombinant adenovirus serotype 5 (rAd5) vectors expressing similar antigens. This approach aims to induce both antibody and T-cell responses against multiple strains of HIV-1. Clinical trials have shown the regimen to be safe and immunogenic—eliciting durable CD4+ and CD8+ T-cell responses as well as binding antibodies—but it did not demonstrate efficacy in preventing HIV infection or reducing viral load set point in large phase IIb/III studies. The primary developer is the Vaccine Research Center at the National Institute of Allergy and Infectious Diseases (NIAID)[2][4][1].
