DNA CON-S env is an experimental DNA vaccine candidate designed for the prevention of HIV-1 infection. It consists of a DNA plasmid encoding a synthetic consensus envelope (env) protein, designated CON-S, which is derived from the alignment of multiple HIV-1 Group M sequences. The consensus approach is intended to address the high genetic variability of HIV-1 by providing an antigen that is more closely related to a broad range of circulating viral strains than any single natural isolate. Developed through a collaboration involving the Duke Human Vaccine Institute and the National Institute of Allergy and Infectious Diseases (NIAID), DNA CON-S env has been evaluated in Phase 1 clinical trials (such as HVTN 083 and HVTN 085) as a priming agent in heterologous prime-boost regimens involving viral vectors like MVA-CMDR or NYVAC.
