Overview

DNA-HIV-PT123 is an experimental DNA plasmid vaccine candidate developed for the prevention of HIV-1 infection. It consists of DNA plasmids encoding specific HIV-1 antigens, typically including Gag, Pol, and Env proteins, designed to stimulate both cellular and humoral immune responses. The vaccine is primarily utilized as a priming agent in heterologous prime-boost regimens, often followed by boosts with viral vector vaccines (such as NYVAC-HIV-PT1 or NYVAC-HIV-PT4) or protein subunit vaccines (such as AIDSVAX B/E). Clinical development has been led by the National Institute of Allergy and Infectious Diseases (NIAID) and the HIV Vaccine Trials Network (HVTN), with Phase 1 and 1b studies evaluating its safety, tolerability, and immunogenicity in healthy, HIV-uninfected adults across various global populations.
