DNA-HIVIS is an experimental multiclade DNA-based HIV-1 vaccine candidate developed for use in prime-boost immunization strategies. It consists of a mixture of seven plasmids encoding HIV-1 genes from subtypes A, B, and C, specifically targeting the gp160 (envelope), gag, pol, and rev proteins. The vaccine is designed to elicit broad cellular and humoral immune responses by introducing these genetic sequences into host cells, which then express the viral antigens to prime the immune system. In clinical trials, such as the HIVIS03 and HIVIS09 studies conducted in Tanzania and Sweden, DNA-HIVIS has been primarily evaluated as a priming agent followed by a booster dose of a recombinant viral vector, such as Modified Vaccinia Ankara (MVA-CMDR). The development of this candidate was a collaborative effort led by researchers at the Karolinska Institutet and the Muhimbili University of Health and Allied Sciences.
