Overview

DNA.HTI is a therapeutic DNA vaccine candidate developed by Aelix Therapeutics for the treatment of HIV-1 infection. The vaccine consists of a DNA plasmid encoding the HIVACAT T-cell Immunogen (HTI), a synthetic polypeptide sequence that incorporates 16 highly conserved regions from the HIV-1 proteome (specifically from Gag, Pol, Vif, and Nef proteins). These regions were selected based on their association with viral control in "HIV controllers"—individuals who naturally maintain low viral loads without medication. By focusing the T-cell response on these vulnerable and conserved epitopes, DNA.HTI aims to prevent viral escape and enable long-term viral suppression in the absence of antiretroviral therapy (ART), a goal known as a functional cure. It is typically administered as part of a heterologous prime-boost regimen, often followed by viral vector-based vaccines such as MVA.HTI or ChAdOx1.HTI.
