DNA.HTI + MVA.HTI is a combination therapeutic HIV vaccine regimen developed to elicit strong and broad HIV-specific T-cell responses in people living with HIV (PWH). The vaccines use the proprietary HTI immunogen sequence, which targets conserved regions of the HIV Gag, Pol, Vif, and Nef proteins associated with viral control in humans[1][7]. The regimen typically involves priming with a DNA-based vector encoding HTI (DNA.HTI), followed by boosting with a modified vaccinia Ankara vector also encoding HTI (MVA.HTI)[5][8]. This prime-boost approach is designed to enhance cellular immune responses—specifically CD4+ and CD8+ T cells—against key regions of the virus[1][5]. Clinical trials have shown that this combination is safe and highly immunogenic in early-treated individuals on antiretroviral therapy (ART), leading to robust increases in HTI-specific T cell frequencies[5][6]. While it does not prevent viral rebound after ART interruption on its own, stronger vaccine-induced immune responses correlate with longer periods off ART before viral rebound occurs[1][6]. The vaccines are being investigated as part of broader "cure" strategies for HIV infection.
