dnaJP1 is a synthetic 15-amino acid peptide derived from the heat shock protein dnaJ of *Escherichia coli*. It specifically contains the "shared epitope" sequence (QKRAA), which is a five-amino acid motif found in the HLA-DRB1 alleles that are strongly associated with increased susceptibility to and severity of rheumatoid arthritis (RA). The drug is developed as an oral tolerogen, a type of immunotherapy designed to "re-educate" the immune system. By delivering the peptide orally, it interacts with the mucosal immune system in the gut to induce peripheral tolerance. This process involves the generation of antigen-specific regulatory T cells (Tregs) that produce anti-inflammatory cytokines like IL-10 and TGF-beta, thereby dampening the autoimmune Th1-mediated inflammatory response against joint tissues in RA patients.
