DNK333 is an orally active small molecule drug developed by Novartis as a dual antagonist of the neurokinin 1 (NK1) and neurokinin 2 (NK2) receptors. It was investigated primarily for the treatment of diarrhea-predominant irritable bowel syndrome (IBS-D), asthma, chronic obstructive pulmonary disease (COPD), rhinitis, atopic dermatitis, pruritus, and cough. The mechanism of action involves blocking both NK1 and NK2 receptors—members of the tachykinin receptor family—which are implicated in visceral hypersensitivity and exaggerated motor/secretory activity in IBS-D as well as bronchoconstriction in respiratory diseases. Clinical studies showed that DNK333 was generally well tolerated with a safety profile similar to placebo; some efficacy signals were observed for IBS-D symptom relief[1][7]. Despite reaching phase II clinical trials across several indications—including IBS-D and respiratory conditions—development has been discontinued for all known indications[2][3][5].
