Overview

DNM0547 is a preclinical **small-molecule antibiotic** being developed by DeNovaMed for topical treatment of **stage 1 and 2 diabetic foot ulcers** and other difficult skin and soft-tissue infections. It is described as a **first-in-class inhibitor of acyl carrier protein synthase**, targeting the bacterial enzyme **ACP synthase** involved in lipid metabolism, a pathway not targeted by current antibiotics. The compound was identified through computer-aided drug design from a library of more than 750 novel chemical entities and has shown activity against multidrug-resistant pathogens including **MRSA**. DeNovaMed reports high skin permeability for DNM0547, supporting its development for localized infections such as cellulitis, skin infections, peripheral arterial disease infections, and ischemic limb infections; the company has also studied a separate combination approach of DNM0547 with colistin for more advanced diabetic foot ulcers.
