Overview

DNSP-11 is a synthetic, amidated 11-amino acid peptide derived from the pro-domain of human glial cell line-derived neurotrophic factor (GDNF). It exhibits **neurotrophic and neuroprotective effects** on dopamine neurons, both in vitro and in vivo. Mechanistically, DNSP-11 promotes neuronal survival, differentiation, and neurite outgrowth, protects dopaminergic neurons from toxin-induced apoptosis, and increases dopamine and dopamine metabolite levels in animal models. Unlike GDNF, DNSP-11 does not act via GFRα1/RET signaling, but instead interacts with various cytosolic metabolic proteins, suggesting a distinct neuroprotective mechanism possibly related to mitochondrial function. It activates ERK1/2 phosphorylation and prevents caspase-dependent cell death. DNSP-11 is being explored as a potential treatment for Parkinson's disease, delivered via intranasal and intracerebral routes, and has shown efficacy in improving behavioral and biochemical markers in rodent models of Parkinson's disease[1][2][4][6][8].
