Overview

DNT-1569 is a highly selective, orally active small-molecule inhibitor of poly(ADP-ribose) polymerase 7 (PARP7) developed by Denovicon Therapeutics for the treatment of solid tumors, including lung, breast, ovarian, head and neck, pancreatic, and colorectal cancers. Designed using Denovicon's physics-based generative AI platform, DNT-1569 acts as an immunomodulator that restores type I interferon signaling in tumor cells, activating the innate immune system to potentially convert immunologically "cold" tumors into "hot" tumors. It exhibits potent PARP7 inhibition (IC50 of 4 nM) with approximately 2,000-fold selectivity over PARP1 and 2,500-fold selectivity over PARP2, a profile designed to avoid the hematological toxicities associated with off-target PARP1/2 inhibition.
