DNT-UHN-1 is an experimental adoptive cell therapy consisting of allogeneic double-negative T (DNT) cells, which are defined as mature T lymphocytes that express the CD3 receptor but lack both CD4 and CD8 co-receptors (CD3+ CD4- CD8-). Developed by the University Health Network (UHN), these cells are isolated from healthy donors and expanded ex vivo using a proprietary manufacturing process. DNT cells possess unique innate-like anti-tumor properties, allowing them to target and eliminate malignant blasts in conditions such as acute myeloid leukemia (AML) through both T-cell receptor (TCR)-mediated and natural killer (NK) cell-like recognition mechanisms. Crucially, allogeneic DNT cells do not induce graft-versus-host disease (GvHD), which facilitates their development as an "off-the-shelf" cellular immunotherapy that does not require stringent HLA matching between donor and recipient.
