Drug intelligence / Profile preview
DNTH312 is an investigational, first-in-class, next-generation bifunctional fusion protein developed by Dianthus Therapeutics for the treatment of severe autoimmune diseases. It is designed to simultaneously inhibit two validated pathways: the classical complement pathway by blocking active C1s (aC1s), and B-cell activity by blocking BAFF (B-cell activating factor) and APRIL (a proliferation-inducing ligand). The molecule combines claseprubart (an active C1s antibody) with TACI (transmembrane activator and CAML interactor) and is enhanced with YTE half-life extension technology to enable infrequent subcutaneous self-administration. DNTH312 is aimed at autoantibody-mediated diseases such as myasthenia gravis (MG), chronic inflammatory demyelinating polyneuropathy (CIDP), and multifocal motor neuropathy (MMN), with plans to expand into other autoimmune and rheumatology indications. Dianthus Therapeutics aims to have DNTH312 Phase 1 ready by the end of 2027.
