DNV001 is a GalNAc-conjugated small interfering RNA (siRNA) therapeutic developed by Hangzhou Dinovate Biotech (also known as Hangzhou Dingle Xinwei Biotechnology) for the management of primary hypercholesterolemia and mixed hyperlipidemia. It operates via the RNA interference (RNAi) pathway to selectively silence the expression of proprotein convertase subtilisin/kexin type 9 (PCSK9) in hepatocytes. By inhibiting the production of PCSK9, DNV001 prevents the PCSK9-mediated degradation of low-density lipoprotein (LDL) receptors, thereby increasing receptor availability on the liver cell surface and enhancing the clearance of LDL cholesterol (LDL-C) from the bloodstream. DNV001 is notable for being produced through a novel chemoenzymatic ligation synthesis process in partnership with Hongene Biotech, which improves purity and scalability compared to traditional solid-phase synthesis. Preclinical and early clinical data suggest that DNV001 provides durable lipid-lowering efficacy, potentially requiring only twice-yearly subcutaneous injections.
