Overview

DNV3681 is a novel, dual-action antibiotic belonging to the oxazolidinone-quinolone hybrid class. It is the active metabolite of the prodrug DNV3837 (formerly MCB3837). DNV3681 exhibits potent activity against Gram-positive bacteria, particularly *Clostridioides difficile*, by simultaneously inhibiting bacterial protein synthesis (via the 50S ribosomal subunit) and DNA replication (via DNA gyrase and topoisomerase IV). This hybrid structure is designed to overcome resistance mechanisms and provide a narrow-spectrum profile suitable for treating recurrent *C. difficile* infections (rCDI) while minimizing disruption to the gut microbiota. The prodrug DNV3837 is administered intravenously and rapidly converted to DNV3681 in the blood, which then reaches the site of infection in the intestine.
