Overview

DNX-03000 is a preclinical-stage small molecule activator of pyruvate kinase M2 (PKM2) developed by Dynamix Pharmaceuticals. PKM2 is a key glycolytic enzyme that is typically found in a low-activity dimeric state in cancer cells, facilitating the diversion of glucose metabolites into biosynthetic pathways to support rapid cell proliferation, a phenomenon known as the Warburg effect. DNX-03000 acts as a positive allosteric modulator, promoting the conversion of PKM2 into its highly active tetrameric form. This metabolic shift increases the flux of glucose toward energy production and reduces the availability of precursors for macromolecular synthesis, thereby exerting anti-tumor effects. The program was primarily targeted at solid tumors, though development appears to have halted following the closure of Dynamix Pharmaceuticals.
