Overview

DNX-1000 is a small molecule inhibitor of Polo-like kinase 1 (PLK1) developed by Dynamix Pharmaceuticals. PLK1 is a serine/threonine kinase that serves as a master regulator of the cell cycle, particularly during the transition into and through mitosis. By selectively inhibiting PLK1, DNX-1000 disrupts centrosome maturation, spindle formation, and chromosome segregation, leading to mitotic catastrophe and apoptosis in cancer cells. The compound is designed to target the high mitotic index of malignant cells while potentially offering a therapeutic window over normal tissues. It is currently in Phase 1 development for the treatment of advanced cancers, including solid tumors and hematological malignancies.
