Overview

DNX-114 is a preclinical, second‑generation, long‑acting, high‑affinity soluble tumor necrosis factor (TNF) receptor–based biologic being developed using PASylation technology to extend plasma half‑life and reduce dosing frequency in chronic inflammatory diseases.[3][5][7][13] Designed as an anti‑cytokine therapeutic targeting TNF, it is being advanced by DNX Biopharmaceuticals in collaboration with XL-protein for immune-mediated conditions such as rheumatoid arthritis, ankylosing spondylitis, and other rheumatic indications where TNF plays a central pathogenic role.[1][3][5][10][13] The molecule incorporates XL-protein’s PASylation platform, a biological alternative to PEGylation, to increase molecular size and circulation time while maintaining high-affinity TNF binding, enabling sustained TNF neutralization for improved control of inflammation in autoimmune and immune system diseases.[3][5][7][13]
