Drug intelligence / Profile preview
DO34 is a highly potent, selective, and centrally active dual inhibitor of diacylglycerol lipases alpha (DAGLa) and beta (DAGLb), which are the primary enzymes responsible for the biosynthesis of the endocannabinoid 2-arachidonoylglycerol (2-AG). Developed by researchers at The Scripps Research Institute and Leiden University, DO34 acts by blocking de novo 2-AG synthesis, thereby suppressing tonic cannabinoid receptor 1 (CB1) activation. It is widely used as a chemical probe in preclinical research to study endocannabinoid signaling, synaptic plasticity, neuroinflammation, pain, and behavioral processes such as fear extinction, fear memory generalization, and alcohol consumption.
