Docetaxel + fluorouracil + leucovorin is a combination chemotherapy regimen used primarily for the treatment of advanced or metastatic gastric cancer. Docetaxel is a taxane that inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis. Fluorouracil (5-FU) is an antimetabolite that inhibits thymidylate synthase and interferes with DNA synthesis. Leucovorin enhances the binding of 5-FU to thymidylate synthase, increasing its cytotoxicity. This combination has demonstrated efficacy in patients with advanced gastric cancer and offers an alternative to cisplatin-containing regimens[1][3][5]. The regimen may be administered as infusional or bolus dosing schedules.
