A three-drug chemotherapy regimen combining the taxane docetaxel, the platinum agent carboplatin, and the oral fluoropyrimidine prodrug capecitabine. Docetaxel stabilizes microtubules and inhibits depolymerization to block mitosis; carboplatin forms DNA intra- and interstrand crosslinks to inhibit DNA replication and transcription; capecitabine is converted to 5-fluorouracil (5-FU), which inhibits thymidylate synthase and incorporates into RNA and DNA, impairing nucleic acid synthesis. The combination has been studied primarily in advanced or metastatic esophagogastric cancers as a modification of docetaxel–platinum–fluoropyrimidine regimens, with varying schedules evaluated in phase I/II and a randomized phase II trial.
