This is a combination therapy regimen consisting of three chemotherapeutic agents—docetaxel, carboplatin, and gemcitabine—administered alongside radiotherapy. Each component has a distinct mechanism of action: - Docetaxel is a taxane that promotes microtubule assembly and inhibits their disassembly, thereby disrupting mitosis and inducing apoptosis in rapidly dividing cells. - Carboplatin is a platinum-based compound that forms DNA crosslinks, inhibiting DNA synthesis and function. - Gemcitabine is an antimetabolite (nucleoside analog) that incorporates into DNA during replication, causing chain termination and cell death. Radiotherapy uses ionizing radiation to damage the DNA of cancer cells directly or indirectly through free radical formation. This combination regimen has been studied primarily for advanced cancers such as non-small cell lung cancer (NSCLC) and urothelial carcinoma in patients who may not be suitable for cisplatin-based regimens[1][2][5]. The combined approach aims to maximize cytotoxic effects on tumor cells by targeting different pathways involved in cell division and survival.
