TPF2 (also referred to as TP²F) is an experimental induction therapy regimen combining the standard-of-care TPF chemotherapy (docetaxel, cisplatin, and 5-fluorouracil) with the immune checkpoint inhibitor pembrolizumab. This combination is being evaluated for the treatment of untreated, locally-advanced, unresectable squamous cell head and neck carcinoma (HNSCC). The regimen aims to leverage the cytotoxic effects of chemotherapy to debulk the tumor and potentially enhance the anti-tumor immune response induced by pembrolizumab, which targets the programmed cell death protein 1 (PD-1) receptor. Following induction with TPF2, patients typically proceed to radiotherapy combined with carboplatin.
