Overview

This combination regimen, evaluated by the University of Alabama at Birmingham in a Phase I/II trial (NCT00318890), is designed for the treatment of locally advanced squamous cell carcinoma of the head and neck (HNSCC). The protocol involves an induction phase using docetaxel (a taxane microtubule stabilizer) and cisplatin (a platinum-based DNA cross-linking agent), followed by a concurrent phase where docetaxel is administered alongside radiotherapy. Subcutaneous amifostine is included as a radioprotectant to mitigate toxicities associated with radiation, such as xerostomia. The regimen aims to improve both local-regional control and reduce systemic metastasis in patients with Stage III or IV disease by combining potent cytotoxic agents with a cytoprotective strategy.
