The TPC regimen is a combination chemotherapy and targeted therapy protocol consisting of docetaxel, a platinum agent (cisplatin or carboplatin), and cetuximab. Developed and evaluated by Stanford University in a Phase 2 clinical trial, this regimen is specifically designed for the palliative treatment of patients with recurrent or metastatic squamous cell carcinoma of the head and neck (SCCHN). The protocol utilizes a weekly administration schedule (3 weeks on, 1 week off) to enhance tolerability while maintaining efficacy compared to standard every-3-week regimens. Docetaxel acts as a microtubule stabilizer, the platinum agents function as DNA cross-linkers, and cetuximab is a monoclonal antibody targeting the epidermal growth factor receptor (EGFR), collectively providing a multi-targeted approach to inhibit tumor growth and progression.
