This combination chemotherapy regimen, often referred to as the DCU regimen, consists of docetaxel, cisplatin, and oral tegafur/uracil (UFUR) supplemented with leucovorin. It was evaluated in a Phase II clinical trial led by Taipei Veterans General Hospital for the treatment of advanced oral tongue squamous cell carcinoma (OTSCC). The strategy employs induction chemotherapy to shrink tumors before tongue conservation surgery, followed by risk-adapted postoperative adjuvant therapy. Docetaxel acts as a microtubule stabilizer, cisplatin as a DNA-damaging agent, and tegafur/uracil as an oral fluoropyrimidine that inhibits thymidylate synthase. This multi-modal cytotoxic approach targets rapidly dividing cancer cells to improve organ preservation and survival outcomes in patients with cT2-4 N0-2 M0 stage disease.
