Docetaxel + diethylstilbestrol is a combination therapy investigated primarily for the treatment of metastatic androgen-independent (castration-resistant) prostate cancer. Docetaxel is a semisynthetic taxane antineoplastic agent that promotes microtubule assembly and stabilizes microtubules by preventing their depolymerization, thereby inhibiting cell division and inducing apoptosis in cancer cells[1][2][6]. Diethylstilbestrol (DES) is a synthetic nonsteroidal estrogen that was historically used for menopausal disorders but has also been studied as an adjunct in prostate cancer due to its hormonal effects[5]. Preclinical studies suggest this combination may have additive or synergistic activity against prostate cancer, with DES potentially inhibiting expression of taxane-resistant tubulin isoforms[3][4]. Clinical trials have shown significant PSA response rates in patients with advanced disease, though toxicity can be substantial[3][4].
