Docetaxel + doxercalciferol is a combination of two drugs: docetaxel, a taxane-class antineoplastic agent, and doxercalciferol, a vitamin D analog. Docetaxel works by promoting the assembly and stabilization of microtubules in cancer cells, thereby inhibiting their normal dynamic reorganization required for cell division and leading to apoptosis. It is used in the treatment of various cancers including breast, lung, prostate, stomach, and head and neck cancers[3][5][6]. Doxercalciferol is an inactive prohormone that undergoes hepatic conversion to active metabolites; it acts as a vitamin D receptor agonist to regulate calcium metabolism and suppress parathyroid hormone production. It is primarily indicated for secondary hyperparathyroidism in chronic kidney disease patients but has also been studied for its potential anti-tumor effects when combined with chemotherapy agents like docetaxel[1][2]. The combination has been evaluated in clinical trials for metastatic androgen-independent prostate cancer (AIPC), where preclinical data suggested synergistic activity[2].
