Each drug has a distinct mechanism of action: - Docetaxel is a taxane antineoplastic agent that promotes microtubule assembly while inhibiting their disassembly, thereby disrupting mitosis and leading to cell death[4][5][8]. - Doxorubicin is an anthracycline antibiotic that intercalates DNA strands and inhibits topoisomerase II, preventing DNA replication and transcription in rapidly dividing cells[6]. - Ifosfamide is an alkylating agent that crosslinks DNA strands, resulting in inhibition of DNA synthesis and function[1][5]. This combination exploits different mechanisms to maximize cytotoxic effects against cancer cells. The regimen is administered intravenously in cycles with supportive medications such as mesna (to prevent bladder toxicity from ifosfamide) and granulocyte colony-stimulating factors (to reduce neutropenia risk)[2]. The combination has demonstrated efficacy particularly in soft tissue sarcoma, advanced urothelial cancer after platinum-based therapy failure, as well as other solid tumors[2][5][8].
